I think Malwarebytes alone can do it, with a scan from CCleaner. You must have some special reason to use Windows Defender I think ?
I run Defender + MBAM as well as an occasional CCLeaner too. That keeps my rigs at home and portable units safe and clean overall. That might be too much for a few and not enough for many though, just depends on habits and what one chooses to get into, chooses what to click on, and where to browse. A lot of issues can be prevented by smart browsing. Another thing to keep in mind that is often ignorantly overlooked is monitoring firewall rules. MS's firewall is good enough when teamed with a decent router + firewall. I wish I had a better router, and would love a Sonicwall or something higher-grade. But managing rules and running consistent scans is the way to go imho.
I prefer MBAM free so I update and scan when I need it...and I utilize Defender/MSE as my automated/live scan utility...for me it seems to consume little resources and doesn't seem to degrade my gaming performance whilst I am still able to keep a clean running system. I try to run an MBAM quick scan (on 1.75, and the standard smart scan on 2.00, depends on what version you like...both are solid imho, I also like 2.0 better). Having other utilites at the ready isn't a bad idea, ADW, JRT, Hitman...things to run if you truly get infected because something snooped into your system and planted something nasty.
I end up dealing with a lot of customers (dozens/week) that run just one solution, even enterprise-grade solutions don't cut it alone. Even for those that keep their nose out of the dark corners of the Internet where they shouldn't be going should they not be seeking infection and risks.
Keeping run-times like Java, Adobe products, etc updated is critical too...I usually run through a ninite run of those once a month too just to ensure they're updated.
It's easy to assume you won't be infected, but pick the utilities you want, keep them updated and keep open to using more utilities, understanding how to manage your firewall, how to find and follow articles to remove the nastier infections and you'll be fine.
