Having experience with both types of setups, the GTX480 will be smoother than the SLI config.

Efficiency of fermi is much better, on gpu cache is much better. Better memory controller and more VRAM. So even with just one card I have seen rigs run so much smoother, I played around alot before the hydro copper came in. GTX 275 SLI and GTX 280 SLI got smoked in real life smoothness.

Fermi drivers helped too the series did very well in my opinion. If you compare GTX 480 to the GTX 570, I would take a 480 any day. The 480's are tough as nails, they might run hot but I have never been seen one go down and pushed some high volt through 3 different ones.
