System Specs

DDR3 1600 or 1333

Is there a big difference? Will I see it?

You will. PC3-12800 will clock down to 1333 and give better timings than 1333. Personally I have PC3-12800 in my machine right now underclocked to 1200MHz but the timings are 5-5-5-10 that's pretty nice for DDR3

Of course. When I upgraded from DDR2 800 to 1066, I didn't notice a difference
