Will 6870 x2 work on MSI P67S-C43?

It says the motherboard doesn't support Crossfire (here) but since 6870 x2 is only 1 board, will it run Crossfire on this motherboard?

yes it will - the 6870x2 is still a single card. despite having 2 GPu cores on it.

Crossfire is traditionally done with 2 graphic cards

Yep. The card has a PLE PCI-E to PCI-E bridge chip on it, so like any other dual GPU card, it CF's on itself, not on your mobo. After seeing this post though (and telling him to not get it before), I'd suggest staying away from that card.

It's a rather cheaply made non-ref board with possible driver issues. You're better off with a single, powerful GPU.
