Is there are memtest program that can check over 2 Gb of single DIMM and over 4 Gb of total memory?
Because i just have the old memtest86, which is limited to 4 Gb.

it should if you go under advanced options. I used a version from like 4 years ago and iv tested over 8GB with it. If it really doesnt work for you the new version should it was updated lat year IIRC

Does memory diag need to be run on default RAM SPD settings, or can i run this program in RAM OC mode? By OC i mean default safe OC, defined by manufacturer (CL 7 at 1.65V versus SPD settigns CL9 at 1.5V)

you can use it at whatever speed you want. its not just used to see if a stick is bad alot of us use it to test ram stability when we OC.
